She attended Senn High School and later studied at the Goodman School of Drama at the Art Institute of Chicago. In 1948, she was crowned Miss Chicago and became a semifinalist in the Miss America pageant, which helped her gain recognition and opened doors for her acting career.

Lois Nettleton began her career in theater, performing in Broadway productions such as "Cat on a Hot Tin Roof" and "A Streetcar Named Desire."
